Detection Method:Sandwich
Test principle:This assay employs a two-site sandwich ELISA to quantitate AP12 in samples. An antibody specific for AP12 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyAP12 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for AP12 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of AP12 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Product Overview:Apelin, the endogenous peptide ligand of the APJ receptor, is expressed in brain regions implicated in food and water intake. The APJ receptor belongs to a family of 7-transmembrane G-protein coupled receptors.The ligand was named apelin after APJ endogenous ligand. Apelin and APJ are widely expressed in homogenates from animal organs in a pattern shared with angiotensinogen and the angiotensin receptor. Apelin is widely distributed in the CNS and periphery, especially in the heart, kidney, lung and mammary glands.present study investigated basal plasma levels of apelin, orexin-A, and leptin in morbid obese patients.Apelin-12 levels also were inversely correlated with levels of nt-proBNP.
